I have a valid copy of Windows XP Home edition on my computer and I bought a Windows XP Pro Upgrade. I want to reformat my computer before installing XP pro, does the upgrade disk need to see that a previous version is installed on the computer?

2 ways to do this. you can boot to the upgrade cd, then insert the xp home cd you're upgrading from when prompted. or you can install xp home fully, then run the xp pro upgrade from your desktop.
Yes the XP pro upgrade disk will allow a clean install.
The better way is to
Start the computer let it boot up with XP home. Put the XP Pro disk in the machine and let it start the upgrade process. Once into the process you will be given to option to delete all partitions and format the computer and continue the XP Pro installation process.
That way you don't have to mess with formatting the hard drive prior to Installing Windows, It will be done as part of the XP Pro installation process.
The other way there is a chance the Computer will not recognize the CD/DVD ROM drive and you will not be able to boot from the XP Pro installation disk… Unlikely it could happen.
