Yes, they can harm ur registry and mess it up, so I advice u to do a backup befor each cleaning process.
The safest registry cleaner ever is a program called VCOM Fix-It Utilities.

I personally have tried most of registry cleaner programs and can confirm about regsupreme, tuneup utilities, fix-it utilities, system mechanic, and norton windoctor……u should avoid regcure, and other crap ones which is really useless and do nothing in the registry but they can also mess it up.
I know many people may recommend ccleaner but this is only because it’s free……this program is fantastic in junk file very cleaner but very poor in registry cleaner.
About Norton systemwork it’s very safe and not that aggreasive registry cleaner!

Not really safe, your computer will crash , freeze up, waste your productive time on scanning for viruses, and on and on. you will need months to learn how to use this complex software. By the time you know how to use it. You already did all the damaged to your computer without knowing the right answer with the pop up alerts. Please do yourself a favour stay with Microsoft’s softwares.
I been using Norton for seven years, And never had a problem since then.
Yes, there’s always that chance. I use Crap Cleaner (CCleaner) I always back a registry backup just in the case that you accidentally delete something critical.
To create a regisrty backup yourself, follow these instructions:
1. Hit windows key +R to open run
2. Type ‘regedit’ no quotes
3. Click "My Computer"
4. File>Export
5. Make sure all is checked.
6. Name it and click save.
It may take a minute or two to save.
