In a sense it does though not greatly. A PC performance is mostly affected by the amount of available memory. Another factor is the CPU speed, one which runs on P3 is slower than one which runs on P4.
Registry clean up will just help you to remove old unused registries as well as repair broken registries.

Not to the point of you saying WOW THAT IS FAST. Over time all computers slow down from adding and deleting files. If you really want to speed up your pc you can
1. restore the whole pc to factory settings
2. to make the boot up faster type msconfig in the run screen or start search if your using vista and go to startup and take off what you do not need
3.if your frisky use Linux it does not have to even defrag it is always running at full optimization.
