Following are the causes of system slow down:
1. File fragmentation
2. Registry data fragmentation
3. A large number of installed programs
4. Disk allocation by unnecessarily stored files
5. A number of items to load at the system startup
Resolution:

[A] Optimize the System Performance:

1. Defragment the disks
2. Defragment the system registry [Using RegInOut]
3. Fix the errors in the system registry [Using RegInOut]
4. Prevent execution of unnecessary startup and services items
5. Delete unnecessary files and stuff
6. Uninstall the programs that you never use
7. Install additional Memory (RAM)
8. Install a Graphics Card
9. Change settings for "Best Performance"; rather than "Best Appearance"

(1) Disabling Unnecessary Add-Ons:
1. Open Internet Explorer.
2. Click Tools | Manage Add-Ons.
3. In the "Show" drop-down box, select "All Add-Ons".
4. Disable those add-ons you no longer needed.
5. Click "Close" and restart the browser.
(2) Clearing Browser Stuff:
1. Download RegInOut.
2. Install it on your computer; and then run it.
3. Click "System Cleaner", and then "Clean" button.
Open Internet Explorer, and click Tools -> Delete Browsing History.

A possibility that exists in regards to your computer slowing down could be the type of antivirus/firewall you’re using. You didn’t mention if you were using Nod32 (ESET), AVG, McAfee, Norton, or any sort of protection. Potentially, if you happen to be using Nod32 or Smart Security by ESET it is a major resource hog. It’ll slow your internet down as well as processes on your computer itself. It depends on what you have and the options enabled.
Also, add Spybot – Search & Destroy to your list of programs because it helps a lot.
Lastly, I’d just take a look at your running processes and see whats eating up the most memory and if there are any you don’t recognize look them up.
I’ve got a four year old computer running XP as well as a two year old laptop running Vista, and neither of them experience lag or ever run slow. You just kind of have to check all the little things as well.
Either a virus (Malwarebytes can miss stuff too), or iTunes 10. Try uninstalling it or rolling back to iTunes9 and see what happens.
Also a good idea is to check running processes and research anything you don’t know on http://www.fileinspect.com
